Mutation details: A version of the open reading frame with exons 7 through 10 replaced by a floxed cDNA fragment encoding amino acids 219 to 376 and neo cassette followed a modified exon 7 with a stop codon and polyadenylation signal attached at the 3' end was knocked into the gene. These modifications would direct the expression of only the 'a' alternative transcript instead of the full range transcipts, 'a' through 'e'. Modifications to the genomic DNA were confirmed by Southern blot analysis and PCR genotyping. The presence of only one splice variant was confirmed by RT-PCR and western blot analysis on proteins extracted from the liver.
