Mutation detailsA targeting construct with the SA-PhiC31opA-PGKneopA cassette, which contains the mouse codon-optimized site-specific recombinase PhiC31o sequence (with a C-terminal SV40 nuclear localization signal) driven by the phosphoglycerate kinase 1 (PGK) promoter, and bovine growth hormone polyadenylation (bpA) sequence was inserted into the Gt(ROSA)26Sor locus. Flp-mediated recombination removes the neo cassette. PhiC31o-mediated recombination in mice carrying containing a sequence of interest flanked by attB and attP sites results in tissue-specific deletion of the targeted sequence. (J:128069)
