Mapping and Phenotype information for this QTL, its variants and associated markersJ:132289Linkage analysis was performed on 224 CAST/EiJ x (CAST/EiJ x C57BL/6J)F1 backcross animals to identify quantitative trait loci (QTL) associated with the entrainment of circadian rhythms. Animals were monitored for wheel running activity for more than 21 days. Parental strain CAST/EiJ displays significantly earlier onset of running activity compared to parental strain C57BL/6J. A panel of 84 polymorphic markers at 20 cM resolution was used for genome scan. Significant linkage to early running activity mapped to 41 cM on mouse Chromosome 18 near D18Mit184 (LOD=3.9-4.0). This locus is named Era1 (early running activity 1). The QTL interval of Era1 spans 31 cM to 50 cM. Animals homozygous for CAST/EiJ-derived alleles at Era1 have a high incidence of early wheel running (85%) compared to heterozygous animals (45%). This locus explains 10% of the phenotypic variance. Era1 did not show linkage to circadian period, but did show linkage to delays in circadian phase induced by light exposure at specific time points after dark-dark cycle. Homozygosity for CAST/EiJ-derived alleles at Era1 confer decreased phase delay after light exposure early in the subjective night period. Grp (40 cM) is a potential candidate gene located near Era1. Epistatic interaction was detected between a locus on mouse Chromosome 16 near D16Mit34 (9.61 cM) and mouse Chromosome 1 near D1Mit145 (89 cM; LOD=4.3). |
