Mutation details: The entire 1.4 Kb cDNA from a human AKR1B1 (hAR) clone with 36 bo 5' and 325 bp 3' untranslated regions was cloned into a vector containing the SV40 splice site and poly(A) sequence. The -341 to +49 region of the mouse Cryaa promoter was inserted at the 5' end of the AKR1B1 sequence and microninjected into fertilized oocytes. Northern blot analysis showed expression of AKR1B1 in the lens, but not in liver, brain, testis, or muscle. Five lines were characterized, CAR222, -223. -435, -440, and -648 with 30, 70, 100, 70, and 5 copies of the transgene. Lines 222 and 648 were used further and entered into the MGI database.
