Mapping and Phenotype information for this QTL, its variants and associated markers

Genetic loci associated with wheel running distance, duration and speed were mapped using a population of 310 (C3H/HeJ x C57L/J)F2 animals. C57L/J is a high activity strain whereas C3H/HeJ is a low activity strain. A panel of 129 SNPs spaced an average of 13.7 cM apart was used for genome scan. Animals were assessed for wheel running duration, speed and distance at 24 hour intervals starting at age 63 days for 21 consecutive days.

Significant linkage to wheel running duration mapped to 10 Mb (7 cM) on mouse Chromosome 13 near rs6329684 with LPR=4.10. This locus explains 6.24% of the phenotypic variance and is named Vactq1 (voluntary activity QTL 1). The QTL interval spans 1 Mb to 15 Mb. Vactq1 also shows significant linkage to wheel running distance (LPR=4.14, 6.36% of the variance) and speed (LPR=3.98, 6.10% of the variance). C57L/J-derived alleles at Vactq1 confer increased activity measures. Haplotype association mapping (HAM) analysis using 27 inbred strains and 636 SNPs confirmed the presence of Vactq1. Suggestive linkage to wheel running duration mapped to a second locus on chromosome 13 (LPR=3.21 at 42.9 Mb), chromosome 5 (LPR=3.21 at 90 Mb) and chromosome 8 (LPR=2.17 at 8.88 Mb).

Significant linkage to wheel running speed mapped to 13.23 Mb (5cM) on mouse Chromosome 9 near rs13480073 with LPR=7.51. This locus is named Vactq2 (voluntary activity QTL 2). The QTL interval spans 1 Mb to 15 Mb and explains 11.27% of the variance. C57L/J-derived alleles at Vactq2 confer increased wheel running speed. This locus was confirmed using HAM analysis. Vactq2 also shows suggestive linkage to wheel running distance. Suggestive linkage to wheel running speed mapped to 27 Mb (6 cM) on mouse Chromosome 12 with LPR=2.07. Other suggestive loci mapped to chromosomes 2 (19 Mb and 109 Mb), 5 (57 Mb), and 8 (50 Mb and 88 Mb).

Epistatic interaction was detected between suggestive wheel running distance loci on chromosome 5 (103 Mb) and chromosome 9 (1 Mb). TThis locus pair accounts for 24% of the phenotypic variance in wheel running distance.
