Linkage analysis was performed on a population of 60 F2 mice derived from BTBR-Lepob and C57BL/6J-Lepob parental strains to identify QTLs associated with 67 liver metabolites (mQTL) and liver mRNA expression (eQTL). F2 animals were phenotyped between13-26 weeks of age. Parental strain BTBR-Lepob is susceptible to diabetes whereas C57BL/6-Lepob is resistant.

A locus on mouse Chromosome 9 near 102.5 Mb shows significant linkage to liver metabolite glx and liver mRNA transcript levels for Slc1a2 and Arg1. The mQTL interval spans approximately 85.1 Mb to 112 Mb. This locus is named Lmq1 (liver metabolism QTL 1). Slc38a3 (107.5 Mb, 63 cM) maps to this region.

An interval on mouse Chromosome 2 between 114 Mb and 178.5 Mb is linked to liver metabolite glx and liver mRNA levels for Agxt, Arg1, Ass1, Ivd and Slc1a2. This locus is named Lmq2 (liver metabolism QTL 2). Ivd (118.6 Mb), Pck1 (172.8 Mb) and Slc1a2 (102.6 Mb) map to this region.

An eQTL for Arg1 and Asl expression mapped to mouse Chromosome1between 13 Mb and 171.5 Mb. This locus is named Lmq3 (liver metabolism QTL 3). Agxt (95 Mb) is located in this region.

Linkage to liver metabolite glx mapped to mouse Chromosome 4 between 3.5 Mb and 150.1 Mb. This locus is named Lmq4 (liver metabolismQTL4) and coincides with an eQTL for Ass1 and Pck1 liver mRNA levels.

On mouse Chromosome 5, linkage to liver metabolite glx overlaps with an eQTL for Ass1 expression at approximately 39.3 Mb - 129 Mb. This locus is named Lmq5 (liver metabolism QTL 5).Asl(130.3 Mb) is located near this interval.

On mouse Chromosome 7, linkage to liver metabolite glx overlapped with an eQTL for Agxt. This locus is named Lmq6 (liver metabolism QTL 6) and spans approximately 13.9 Mb to 35.1 Mb.

An eQTL for Ass1, Ivd andSlc38a3 liver mRNA levels mapped to mouse Chromosome 8 between 36.6 Mb and 170 Mb. This locus is named Lmq7 (liver metabolism QTL 7).

An eQTL for Pck1 liver mRNA expression mapped to mouse Chromosome 10 between 107.2 Mb and 120.4 Mb. This locus is namedLmq8(liver metabolism QTL 8).

Linkage to liver metabolite glx overlaps with an eQTL for Ivd liver mRNA expression on mouse Chromosome 13 at around 81 Mb. This locus is named Lmq9 (liver metabolism QTL 9).

On mouse Chromosome 14, an eQTL for Agxt and Pck1liver mRNA expression overlap at approximately 21 Mb. This locus is named Lmq10 (liver metabolism QTL 10).

Linkage to Asl and Ass1 liver mRNA expression mapped to 3.3 Mb - 91 Mb on mouse Chromosome 15. This locus is named Lmq11 (liver metabolism QTL11).

Onmouse Chromosome 18, an eQTL for Pck1 and Slc1a2 liver mRNA expression mapped between 15.5 Mb and 85.7 Mb. This locus is named Lmq12 (liver metabolism QTL 12).
