Mutation detailsThrough homologous recombination a modified version of the vector used to create Ightm2Cgn replaced the endogenous sequence. This vector replaces a 3' Igh-D element (DQ52) and Igh-J elements with a recombined antibody variable region derived from a 4-hydroxy-3-nitrophenyl acetyl binding antibody (B1-8). The variable region, a leader sequence that includes a termination codon, and the endogenous IgH promoter were placed in an inverted fashion so that the IgH promoter was adjacent to the intronic enhancer that lies upstream of the heavy chain constant gene segments.
