Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 1, seven pleiotropic skeletal traits QTLs were mapped to 12 cM (Plskt1; LPR=7.74), 27.6 cM (Plskt2; LPR=20.9), 43 cM (Plstk3; LPR=6.78), 47 cM (Plskt4; LPR=18.46), 57.2 cM (Plstk5; LPR=24.16), 88 cM (Plskt6; LPR=10.58), and 110 cM (Plskt7; LPR=6.3).

Plskt1 (pleiotropic skeletal traits 1) is linked to hindlimb obturator foramen maximum length and mandible square root of angular process area. Theconfidence interval for this QTL spans 9 cM to 29 cM.

Plskt2 (pleiotropic skeletal traits 2) is linked to 14 skeletal traits (face width, ilium length, innominate length, ulna length, sacrum width, necropsy weight, zygomatic width, tail length, tibia length, femur length, humerus length, infraspinous length, scapular width and third metatarsal length). The confidence interval for Plskt2 spans 26 cM to 34 cM.

The Plskt3 (pleiotropic skeletal traits 3) interval spans 30 cM to 49 cM. This QTL is linked to seven skeletal traits- eighth thoracic length 1 in females, eighth thoracic length 2, zygomatic length, iliac crest length in females, mandible square root of incisor alveolus area, infraspinous length and scapula spine length.

Plskt4 (pleiotropic skeletal traits 4) is linked to six skeletal traits (ischium length, palate length, ulna length, face height in males, third thoracic length and weight at 10-weeks of age). The QTL interval spans 44 cM to 53 cM.

Plskt5 (pleiotropic skeletal traits 5) is linked to fifteen skeletal traits (face width, mandible square root of masseteric area, mandible square root of condyloid process area, fourth lumbar length, cranial vault width, face length, third caudal lengths 1 and 2, calcaneus length, eighth thoracic length, humerus length, tibia length, sacrum length, hindlimb obturator foramen maximum length and third metatarsal length). The Plskt5 QTL interval spans 55 cM to 59 cM.

The QTL interval for Plskt6 (pleiotropic skeletal traits 6) spans 81 cM to 95 cM. Plskt6 is associated with five skeletal traits (mandible square root of molar alveolus area in males, eighth thoracic length in males, first lumbar length, zygomatic width and cranial vault length).

Plskt7 (pleiotropic skeletal traits 7) is linked to fourskeletal traits (sacrum width, pubic symphysis length in females, skull frontal bone length in females and fourth lumbar length). The QTL interval spans 106 cM to 118 cM.
