Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 2, five pleiotropic skeletal traits were identified at 41 cM (Plskt8; LPR=10.38), 54.7 cM (Plskt9; LPR=11.18), 65 cM (Plskt10; LPR=10.75), 90.6 cM (Plskt11; LPR=8.7) and 97.6 cM (Plskt12; LPR=9).

A single trait QTL for interorbital width named Iowd mapped to 7 cM (LPR=4.44). The Iowd confidence interval spans 0 cM to 15 cM.

Plskt8 (pleiotropic skeletal traits 8) is linked to 2 skeletal traits- third caudal length and mandible square root of angular process area. The QTL interval spans 37 cM to 44 cM.

Plskt9 (pleiotropic skeletal traits 9) is linked to thirty skeletal traits (face height, third thoracic length, zygomatic length, face width, infraspinous length 1 and 2, basicranial length, femur length, humerus length, sacroilium joint length, ulna length, first lumbar length, first lumbar length 1 and 2, fourth lumbar length 1 and 2, ilium length, zygomatic length in males, ischaic-pubic ramuslength in males, palate length in males, necropsy weight in males, fourth lumbar length in males, weight at 10-weeks of age in males, eight thoracic centrum length in males, eight thoracic length 1 and 3, facial length, hindlimb innominate length, sacrallength and third thoracic length 3). The Plskt9 confidence interval spans 51 cM to 57 cM.

Plskt10 (pleiotropic skeletal traits 10) is linked to ten skeletal traits (third caudal length 3 in females, tail length, superior spinous length 1, scapula spine length, third thoracic length 2 in males, sacral width, third caudal length 1 in males, tibia length, third caudal length 2 and pubis length in males). The confidence interval for Plskt10 spans 63 cM to 76 cM.

Plskt11 (pleiotropic skeletal traits 11) spans88 cM to 93 cM and is associated with three skeletal traits- cranial vault length, pubic symphysis length in males and calcaneus length.

Plskt12 (pleiotropic skeletal traits 12) is linked to five skeletal traits (skull frontal bone length in females, iliac crest length, scapular width, mandible square root of condyloid process area in males and fourth lumbar length 3 in males). The QTL confidence interval spans 87 cM to 107 cM.
