Mutation detailsA transgenic construct with the coding sequence of the iCre recombinase followed by the bovine GH polyadenylation signal and a frt-flanked ampicillin cassette was fused to sequences from the proximal promoter and first intron of the mouse thyroglobulin (Tg) gene. The whole construct was introduced into the PAC encoding the Tg gene and by homologous recombination, the ATG of the iCre replaced the ATG of the thryoglobulin gene. The ampicillin cassette was removed by Flp-mediated recombination. Cre activity is observed in nearly all thyrocytes, but not in other cells of the thryoid gland after E10.5. (J:127484)
