Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 3, six pleiotropic skeletal traits QTLs were mapped to 14 cM (Pbln; LPR=5.67), 19.7 cM (Plskt13; LPR=20.15), 40 cM (Plskt14; LPR=20.80), 49 cM (Plskt15; LPR=28.64), 66 cM (Plskt16; LPR=16.15), and 100.8 cM (Ethl; LPR=2.75).

Pbln (pubic bone length) maps to 14 cM on mouse Chromosome 3. This locus has an effect only in female animals. The Pbln QTL interval spans 9 cM to 20 cM.

Plskt13 (pleiotropic skeletal traits 13) displays linkage to two skeletal traits (third caudal length and face width) at 19.7 cM. The QTL interval spans 17 cM to 61 cM.

Plskt14 (pleiotropic skeletal traits 14) is associated with twenty-nine skeletal traits- cranial vaultlength, cranial vault width, forelimb infraspinous length, zygomatic length, face length, mandible square root of angular process area, hindlimb innominate length, scapular width, ischium length, third thoracic length, palate length, square root of incisoralveolus area, tail length, fourth lumbar length 3, basicranial length, humerus length, scapula spine length, tibia length, sacrum length, forelimb superior spinous length 1 and 2, ulna length, femur length, mandible square root of coronoid process area,third caudal length 3, calcaneous length, mandible square root of corpus area, square root of molar alveolus area and third thoracic length. The Plskt14 interval spans 38 cM to 44 cM.

Plskt15 (pleotropic skeletal traits 15) displays peak linkage to ten skeletal traits at 49 cM and the QTL interval spans 46 cM to 53 cM. Plskt15 is associated with eighth thoracic length 1 and 2 in females, face width, ilium length, mandible square root of masseteric area, zygomatic width, sacral width, interorbital width,hindlimb obturator foramen maximum length and fourth lumbar length.

Plskt16 (pleiotropic skeletal traits 16) is associated with nine skeletal traits at 66 cM (pubis length in males, forelimb infraspinous length 2, scapular width, sacroilium joint length, fourthlumbar length 3, ischiac-pubic ramus length, basicranial length, third thoracic length 2 and face height in males). This QTL interval spans 61 cM to 72 cM.

Ethl (eighth thoracic length) exhibits peak linkage in female animals at 100.8 cM. The QTL interval spans 88 cM to 100 cM.
