Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 4, seven pleiotropic skeletal traits were identified at 0 cM (Cvwd; LPR=4.93), 18.1 cM (Plskt17; LPR=6.58), 34 cM (Plskt18; LPR=17.49), 41 cM (Plskt19; LPR=18.16), 50 cM (Plskt20; LPR=23.19), 55 cM (Plskt21; LPR=13.48), and 68.7 cM (Plskt22; LPR=16.97).

Cvwd (cranial vault width) maps to 0 cM on mouse Chromosome 4. The effect of this QTL is seen only in males. The Cvwd interval spans 0 cM to 22 cM.

Plskt17 (pleiotropic skeletal traits 17) maps to 18.1 cM and is linked to five skeletal traits (face width, forelimb infraspinous length 2, eighth thoracic length 1, zygomatic width and humerus length). The QTL interval spans 11 cM to 22 cM.

The QTL interval for Plskt18 (pleiotropic skeletal traits 18) spans 29 cM to 38 cM. This locus is associated with twenty skeletal traits (femur length, hindlimb ischium length, scapular width, mandible square root of corpus area, square root of incisor alveolusarea and square root of molar alveolus area, eighth thoracic length 2, sacral length, fourth lumbar length 1 and 2, hindlimb ischaic-pubic ramus length, hindlimb obturator foramen and innominate lengths, first lumbar length 1, tibia length, ulna length, weight at 10 weeks of age, fourth lumbar centrum height in females, weight at necropsy and third metatarsal length in females). Linkage to these traits peaks at 34 cM.

Plskt19 (pleiotropic skeletal traits 19) maps to 41 cM and is associated with seven skeletal traits (sacroilium joint length, first lumbar length 1, forelimb infraspinous lengths 1 and 2, mandible square root of condyloid process area, square root of molar alveolar area and eighth thoracic length in females). The Plskt19 interval spans 38 cMto 44 cM.

The Plskt20 (pleiotropic skeletal traits 20) QTL interval spans 48 cM to 53 cM and the locus is associated with seven skeletal traits (face length, palate length, scapula spine length, zygomatic length, mandible square root of condyloid process area, interorbital width and forelimb infraspinous length 1). Linkage to these traits peaks at 50 cM.

Plskt21 (pleiotropic skeletal traits 21) maps to 55 cM and is associated with six skeletal traits (sacrum width, basicranial length, eighth thoraciclength 3, cranial vault length, hindlimb iliac crest length in males and third caudal length 3). The Plskt21 QTL interval spans 53 cM to 59 cM.

Plskt22 (pleiotropic skeletal traits 22) maps to 68.7 cM and is linked to eleven skeletal traits (femur length, tail length, third caudal centrumheight, third caudal length 1, third thoracic length 3, hindlimb ilium length in males, tibia length, fourth lumbar length in males, interorbital width, third metatarsal length in males and mandible square root of masseteric area). The QTL interval spans65 cM to 72 cM.
