Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 5, six skeletal trait QTLs were identified at 0 cM (Plskt23; LPR=2.9), 36 cM (Ilcl; LPR=3.07), 53.9 cM (Plkst24; LPR=5.27), 68 cM (Plskt25; LPR=5.9), 76 cM (Plskt26; LPR=10.9), and 104.2 cM (Plskt27; LPR=2.89).

Plskt23 (pleiotropic skeletal traits 23) mapped to 0 cM and is associated with two skeletal traits in female animals (basicranial length and eighth thoracic length 3). The QTL interval spans 0 cM to 8 cM.

A locus associated with hindlimb iliac crest length, Ilcl, mapped to 36 cM. The QTL interval spans 30 cM to 46 cM.

Plskt24 (pleiotropic skeletal traits 24) at 53.9 cM is associated with six skeletal traits (face width in females, first lumbar length 3, hindlimb ilium length and innominate length in males, interorbital width and scapula spine length in males). The QTL interval spans 46 cM to 57 cM.

The QTL interval for Plskt25 (pleiotropic skeletal traits 25) spans 63 cM to 74 cM. This locus is associated with two skeletal traits (eighth thoracic centrum height and third thoracic length 2) with peak linkage at 68 cM.

Plskt26 (pleiotropic skeletal traits 26) mapped to 76 cM and is linked to five skeletal traits (palate length in males, pubis length in females, forelimb superior spinous length 1, hindlimb obturator foramen length and scapular width in males). The Plskt26 QTL interval spans 72 cM - 87 cM.

Plskt27 (pleiotropic skeletal traits 27) mapped to 104.2 cM and is associated with four skeletal traits (mandible square root of coronoid process area, humerus length in males, zygomatic width in males and femur length in males). The QTL interval spans 80 cM to 110 cM.
