Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTL was 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 8, seven skeletal trait QTLs were identified at 7 cM (Plskt28; LPR=8.05), 35 cM (Plskt29; LPR=15.98), 41.7 cM (Plskt30; LPR=13.2), 55 cM (Plskt31; LPR=12.57), 65.8 cM (Plskt32; LPR=11.29), 79 cM (Plskt33; LPR=23.55), and 83.2 cM (Plskt34; LPR=13.1).

Plstk28 (pleiotropic skeletal traits 28) displays peak linkage to five skeletal traits at 7 cM. The QTL interval spans 0 cM to 32 cM. Plskt28 is linked to sacral width, face length, mandible square root of molar alveolus area, first lumbar length 3 and face width.

Plskt29 (pleiotropic skeletal traits 29) is linked to sixteen skeletal traits at 35 cM (eighth thoracic length 2 and 3, eighth thoraciccentrum height in males, hindlimb ischium length and obturator foramen maximum length, fouth lumbar length, mandible square root of condyloid process area, cranial vault height, necropsy weight, weight at 10 weeks of age, cranial vault width, face height, palate length, forelimb superior spinous length 2 and zygomatic width). The Plsk30 interval extends from 27 cM to 36 cM.

The Plskt30 (pleiotropic skeletal traits 30) QTL interval spans 39 cM to 46 cM. This locus linked to three skeletal traits (zygomatic length, scapular width and fourth lumbar centrum height) with peak linkage at 41.7 cM.

Plskt31 (pleiotropic skeletal traits 31) is associated with nine skeletal traits at 55 cM. These traits include third thoracic centrum height, third thoracic length1, 2 and 3, ulna length, ischaic-pubis ramus length, tail length and eighth thoracic length in males. The QTL interval spans 64 cM to 67 cM.

Plskt32 (pleiotropic skeletal traits 32) displays peak linkage to eight skeletal traits at 65.8 cM (third caudallength 2, fourth lumbar length, basicranial length, cranial vault length, sacrum length, eighth thoracic length, sacroilium joint length and mandible square root of angular process area). The QTL interval spans 64 cM to 67 cM.

Plskt33 (pleiotropic skeletal traits 33) is associated with twenty-six skeletal traits and displays peak linkage at 79 cM. The QTL confidence interval spans 73 cM to 82 cM. Skeletal traits affected by Plskt33 include hindlimb calcaneus length in males, third metatarsal length, first lumbar length, eighth thoracic centrum height in males, facial length, forelimb superior spinous length, mandible square root of condyloid process area and square root of molar alveolus area, ulna length, infraspinous length, fourth lumbar length and fourth lumbar centrum length, femur length, ilium length, obturator foramen length, scapula spine length, tibia length, hindlimb innominate length, zygomatic width, first lumbar length and centrum height, humerus length and interorbital width in males.

Plskt34 (pleiotropic skeletal traits 34) displays peak linkage at 83.2 cM and the QTL interval spans 80 cM to 89 cM. This locus is associated with fourteen skeletal traits, including pubis length in females, first lumbar length, third thoracic length, eighth thoracic length in females, skull frontal bone length, mandible square root of masseteric area and square root of coronoid process area, weight at 10 weeks of age, necropsy weight, forelimb superior spinous length, hindlimb ischium length, cranial vault width, scapular width and cranial vault length in females.
