Mapping and Phenotype information for this QTL, its variants and associated markersJ:134690Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements. On mouse Chromosome 7, eight skeletal trait QTLs were identified at 3 cM (Cvwl; LPR=2.71), 29 cM (Plskt35; LPR=10.59), 42 cM (Plskt36; LPR=8.3), 49 cM (Wght; LPR=17.14), 56 cM (Plskt37; LPR=23.56), 63.7 cM (Plskt38; LPR=13.31), 74 cM (Plskt39; LPR=12.66) and 102.7 cM (Sclw; LPR=3.37).Cvwl (cranial vault length) displays peak linkage at 3 cM and the QTL interval spans 0 cM to 21 cM. This locus affects cranial vaultlength in female animals. Plskt35 (pleiotropic skeletal traits 35) is associated with five skeletal traits at 29 cM. The QTL interval spans 24 cM to 46 cM. Skeletal traits linked to Plskt35 include interorbital width, ulna length, hindlimb innominate length in males, tibia length and ilium length in males. Plskt36 (pleiotropic skeletal traits 36) maps to 42 cM and is associated with three skeletal traits (pubis length, first thoracic centrum height and fourth lumbar length 3). The QTL interval spans35 cM to 46 cM.Body weight at 10 weeks of age is linked to 49 cM on chromosome 7. This QTL is named Weigh (weight at 10 weeks of age) and the confidence interval spans 43 cM to 51 cM.The QTL interval for Plskt37 (pleiotropic skeletal traits 37) spans 52 cM to 58 cM. This locus is associated with twenty skeletal traits including eighth thoracic length, cranial vault width, femur length, hindlimb innominate length in females, hindlimb ischium length, scapular width, third caudal width, third thoracic centrum height, zygomaticwidth, forelimb infraspinous length in females, palate length in females, necropsy weight, mandible square root of angular process area in females, basicranial length and scapula spine length. Peak linkage for Plskt37 occurs at 56 cM.Plskt38 (pleiotropic skeletal traits 38) is associated with nine skeletal traits at 63.7 cM. The QTL interval spans 61 cM to 69 cM. Skeletal traits linked to this locus include sacral length, infraspinous length, mandible square root of masseteric area in males, fourth lumbar length, ischaic-pubis ramus length, forelimb superior spinous length and hindlimb ilium length and innominate length in females. Plskt39 (pleiotropic skeletal traits 39) is associated with thirteen skeletal traits with peak linkage at 74 cM. The QTL interval spans 70 cM to 78 cM. Traits associated with Plskt39 include fourth lumbar length, first lumbar length, third caudal length, mandible square root of masseteric area and square root of corpus area in females, eighth thoracic length, weight at 10 weeksof age, sacroilium joint length, cranial vault height, third caudal centrum height in females and zygomatic length in females. Sacral width displays peak linkage at 102.7 cM on chromosome 7. This QTL is named Sclw (sacral width) and the confidence interval spans 92 cM to 113 cM. |
