Mutation details: A 6.6 fragmnent containing the C region of the mouse gamma 2b (Igh-3) gene from clone VD93 (which shows antibody activity to Pseudomonas aeruginosa) and membrane genomic sequences from clone pgamma2b was subcloned into the pUC13 vector (pCgamma2b-mb). The 4.5 kb fragment of containing the VH region of clone VD93 was cloned into pCgamma2b-mb. A clone with proper orientation of Vpseu-Cgamma2b-mb (clone pVCM) was identified. An 11.2 kb fragment from pVCM containing the Vpseu-Cgamma2b-mb region and 150 bp of vector sequence was used to produce transgenic mice of a (C57BL/6 x SJL)F2 background. Several lines were generated with 2 lines, 343-1 and 348-4, being studied in detail. These lines have 16 and 150 copies of the transgene respectively.

Transgenic lines were derived from two transgenic founder mice and designated line 343-1 and line 348A (copy numbers 16 and 150 respectively). Both lines exhibit a decrease in pre-B cell numbers, however, line 343-1 was characterized in more detail.
