Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 13, five skeletal trait QTLs mapped to 6.8 cM (Plskt64; LPR=6.97), 24 cM (Plskt65; LPR=13.76), 31 cM (Plskt66; LPR=17.15), 41 cM (Plskt67; LPR=14.24) and 61 cM (Plskt68; LPR=24.48).

Plskt64 (pleiotropic skeletal traits 64) maps to 6.8 cM and displays linkage to mandible square root of angular process area and hindlimb iliac crest length. The QTL confidence interval spans 3 cM to 8 cM.

Plskt65 (pleiotropic skeletal traits 65) maps to 24 cM and displays linkage to eighteen skeletal traits. The QTL confidence interval for Plskt65 spans 22 cM to 27 cM. Skeletal traits associated with Plskt65 include ischaic-pubis ramus length, humerus length, basicranial length, fourth lumbar length, femur length, hindlimb innominate length, ischium length, sacroilial joint length, tibia length, first lumbar length, ilium length, third caudal length, fourth lumbar length and third metatarsal length.

Plskt66 (pleiotropic skeletal traits 66) maps to 31 cM and displays linkage to eight skeletal traits including third thoracic length, hindlimb obturator foramen length, scapular width, sacral length, scapula spine length, eighth thoracic length, pubis length and mandible square root of coronoid process area. The QTL confidence interval spans 28 cM to 35 cM

A locus at 41 cM displays linkage to five skeletal traits and is named Plskt67 (pleiotropic skeletal traits 67). Associated skeletal traits include forelimb infraspinous length, first and fourth lumbar lengths, third thoracic length and forelimb superior spinous length. The confidence interval for Plskt67 spans 38 cM to 44 cM.

Twenty-eight skeletal traits display linkage to a locus at 61 cM named Plskt68 (pleiotropic skeletal traits 68). The QTL confidence interval spans 57 cM to 65 cM. Skeletal traits linked to Plskt68 include cranial vault length in females, third metatarsal length, zygomatic length and width, ischium length, palate length, basicranial length, face length and width, third thoracic length, hindlimb calcaneus length and innominate length, third caudal length, humerus length, ulna length, forelimb superior spinous length and infraspinous length, tail length, mandible square root of incisor alveolus area in males, mandible square root of masseteric area, tibia length, interorbital width, body weight at 10 weeks of age, body weight at necropsy, femur length, scapula spine length, scapular width and hindlimb obturator foramen length.
