Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 15, four skeletal trait QTLs mapped to 28.1 cM (Plskt75; LPR=5.72), 38 cM (Plskt76; LPR=13.3), 50.5 cM (Plskt77; LPR=5.53) and 59 cM (Plskt78; LPR=5.92).

Plskt75 (pleiotropic skeletal traits 75) maps to 28.1 cM and is linked to seventeen skeletal measurements. The QTL confidence interval spans 17 cM to 40 cM. Skeletal traits associated with this locus include skull frontal bone length in females,mandible square root of angular process area and square root of masseteric area, cranial vault width, forelimb infraspinous length, scapular width, first lumbar length, body weight at 10 weeks of age, necropsy body weight, face length in males, hindlimbcalcaneus length, forelimb superior spinous length, mandible square root of condyloid process area and square root of molar alveolus area and snout length.

Six skeletal traits exhibit linkage to a locus at 38 cM named Plskt76 (pleiotropic skeletal traits 76). The QTL confidence interval spans 33 cM to 43 cM. Skeletal measurements linked to Plskt76 include eighth thoracic length, hindlimb obturator foramen length, mandible square root of condyloid process area, face width in males and ulna length.

Plskt77(pleiotropic skeletal traits 77) maps to 50.5 cM and displays linkage to eight skeletal traits (cranial vault width, palate length, zygomatic width, eighth thoracic length, tibia length, third metatarsal length, first lumbar length and third thoracic length). The Plskt77 QTL interval spans 40 cM to 57 cM.

Plskt78 (pleiotropic skeletal traits 78) maps to 59 cM and is associated with ten skeletal traits. The QTL confidence interval spans 52 cM to 67 cM. Several skeletal traits are linked to this locus inmales including basicranial length, femur length, humerus length, iliac crest length, ilium length, innominate length, zygomatic length and face length. Third thoracic length and mandible square root of coronoid process area are linked to Plskt78 in bothsexes.
