Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 16, three skeletal traits QTLs were identified at 0 cM (Plskt79; LPR=3.3), 24 cM (Plskt80; LPR=10.1) and 39 cM (Plskt81; LPR=8.3).

Plskt79 (pleiotropic skeletal traits 79) maps to 0 cM and is associated with five skeletal traits in males only: face height, palate length, mandible square root of condyloid process area, ulna length and zygomatic width. The QTL confidence interval spans 0 cM to 5 cM.

A locus at 24 cM shows linkage to twenty-four skeletal traits and is designated Plskt80 (pleiotropic skeletal traits 80). The QTL confidence interval spans 18 cM to 29 cM. Skeletal traits associated with Plskt80 include cranial vault height and width,eighth and third thoracic lengths, calcaneus length, body weight at 10 weeks of age, mandible square root of masseteric area and square root of condyloid process area. Plskt80 is associated with the following skeletal traits in males- forelimb infraspinous length and superior spinous length, sacroilium joint length, scapular width, mandible square root of molar alveolus area and square root of incisor alveolus area, hindlimb innmominate length and ilum length. Plskt80 is associated with hindlimb obturatorforamen length and sacral width in females.

Plskt81 (pleiotropic skeletal traits 81) maps to 39 cM and displays linkage to six skeletal traits including hindlimb ischium length, mandible square root of masseteric area, face width, tail length in females, interorbital width and sacroilium joint length. The QTL confidence interval for Plskt81 spans 34 cM to 49 cM.
