Mutation details: A recombined human mu heavy chain immunoglobulin gene was used in the construction of a transgene. Downstream of the last mu constant region exon, two deletions eliminated the cleavage /poly(A) addition signal so that only the membrane bound form IgM would be expressed. Two transgene lines were created, SA and SD, with the SA line having higher expression. Transgene expression was detected by RT-PCR in the spleen and brain of both transgenic lines.
