Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 95 female animals from a (BALB/cJ x C57BL/6J)F2 intercross to identify QTL associated with resistance to Yersinia pestis. A panel of 58 polymorphic markers spaced 15 cM - 30 cM apart was used for the genome scan. Female F2 animals were infected with Y. pestis pgm mutant strain K15 at various doses and evaluated for survival. Parental strain BALB/cJ is highly resistant to Y. pestis infection with a LD50 dosage greater than 250 times the LD50 dosage of C57BL/6J. The early infection bacterial load (24-48 hours post-infection) in liver and spleen is significantly higher in C57BL/6J compared to BALB/cJ. After infection, susceptible strain C57BL/6J also displays suppurative splenitis, suppurative hepatitis, and increased serumIL-6 levels compared to resistant strain BALB/cJ.

Significant linkage to resistance to Y. pestis mapped to 22 cM on mouse Chromosome 17 with LRS=19.1 at D17Mit66 near the H2 locus. This locus is named Plgr1 (plague resistance locus 1). A population of 95 female (BALB/cJ x C57BL/6J)F1 x C57BL/6J backcross animals confirmed linkage to Y. pestis resistance between 24 cM and 30 cM. Authors explain the shift in linkage peak is due to lack of recombination events between 24 cM and 30 cM in backcross mice. BALB/cJ-derived alleles at Plgr1 confer resistance to Y. pestis with a semidominant mode of inheritance.

A congenic line was constructed by transferring BALB/cJ-derived DNA from the first 45 cM of chromosome 17 containing Plgr1 onto a C57BL/6J genetic background (B6.C-Plgr1BALB/cJ). Congenic animals homozygous or heterozygous for BALB/cJ alleles at Plgr1 displayed decreased bacterial titers after infection with Y. pestis similar to the levels seen in resistant strain BALB/cJ.

Suggestive linkage to Y. pestis resistance mapped to 48.1 cM on mouse Chromosome 2 near D2Mit94 (P=0.154) and 22 cM on mouse Chromosome 12 near D12Mit112 (P=0.373). C57BL/6J-derived alleles at D2Mit94 confer resistance to Y. pestis and this locus explains 9% of the phenotypic variance. The chromosome 12 locus explains 7% of the variance.
