Linkage analysis was performed using a backcross derived from closely related inbred strains DBA/1J and DBA/2J. The genetic composition of these 2 inbred strains differs by just 5.6% at the SNP level. A total of 294 mice from (DBA/1J x DBA/2J)F1 x DBA/1Jand (DBA/1J x DBA/2J)F1 x DBA/2J populations were screened for 6,949 polymorphic SNP markers. Backcross mice were placed on a high-fat diet at 8 weeks of age and phenotyped for plasma triglyceride and HDL concentrations at 8, 11 and 16 weeks of age. Parental strain DBA/2J displays significantly increased plasma triglyceride concentrations on a CHOW diet compared to parental strain DBA/1J. Male DBA/2J animals display significantly increased plasma triglyceride and HDL concentrations after an 8 week high fat diet compared to DBA/1J males.

On mouse Chromosome 2, linkage to plasma triglyceride concentration after 3 weeks on a high fat diet mapped to 3 cM near rs13476339 (8.24 Mb) with LOD=4.5 and is named Tgq10 (triglyceride QTL 10). Linkage to triglyceridelevels after 8 weeks of high fat diet mapped to 27 cM (also near rs13476339) with LOD=5.1 and is named Tgq11 (triglyceride QTL 11). DBA/2J-derived alleles at Tgq10 and Tgq11 confer increased triglyceride concentrations. The confidence interval for Tgq10 and Tgq11 both span the same region between 0 cM and 30 cM.

Suggestive linkage to triglycerides on a CHOW diet mapped to 75 cM (LOD=2.9) and is named Tgq12 (triglyceride QTL 12). This QTL has also been detected in previous crosses. [Table 3].
