Mapping and Phenotype information for this QTL, its variants and associated markersJ:142770Linkage analysis was performed using a backcross derived from closely related inbred strains DBA/1J and DBA/2J. The genetic composition of these 2 inbred strains differs by just 5.6% at the SNP level. A total of 294 mice from (DBA/1J x DBA/2J)F1 x DBA/1Jand (DBA/1J x DBA/2J)F1 x DBA/2J populations were screened for 6,949 polymorphic SNP markers. Backcross mice were placed on a high-fat diet at 8 weeks of age and phenotyped for plasma triglyceride and HDL concentrations at 8, 11 and 16 weeks of age. Parental strain DBA/2J displays significantly increased plasma triglyceride concentrations on a CHOW diet compared to parental strain DBA/1J. Male DBA/2J animals display significantly increased plasma triglyceride and HDL concentrations after an 8 week high fat diet compared to DBA/1J males. On mouse Chromosome 10 two QTLs for plasma triglyceride concentration were detected at 52 cM (LOD=4.7) and 60 cM (LOD=5.5). [Table 3].The locus at 52 cM at rs13480749 (105.43 Mb), designated Tgq20, is linked to triglyceride levels on a CHOW diet. The locus at 60 cM at rs3653850 (120.71 Mb), designated Tgq21, is linked to triglycerides in males after 8 weeks on a high fat diet. DBA/2J-derived alleles at Tgq20 and Tgq21 confer increased plasma triglyceride levels. The Tgq20 confidence interval spans 42 cM to 62 cM. The Tgq21 confidence interval spans 37 cM to 62 cM. |
