Tgq22 has a stronger effect in males compared to females.
Mapping and Phenotype information for this QTL, its variants and associated markersJ:142770Linkage analysis was performed using a backcross derived from closely related inbred strains DBA/1J and DBA/2J. The genetic composition of these 2 inbred strains differs by just 5.6% at the SNP level. A total of 294 mice from (DBA/1J x DBA/2J)F1 x DBA/1J and (DBA/1J x DBA/2J)F1 x DBA/2J populations were screened for 6,949 polymorphic SNP markers. Backcross mice were placed on a high-fat diet at 8 weeks of age and phenotyped for plasma triglyceride and HDL concentrations at 8, 11 and 16 weeks of age. Parental strain DBA/2J displays significantly increased plasma triglyceride concentrations on a CHOW diet compared to parental strain DBA/1J. Male DBA/2J animals display significantly increased plasma triglyceride and HDL concentrations after an 8 week high fat diet compared to DBA/1J males. Linkage to plasma triglyceride concentrations on a CHOW diet mapped to 25 cM on mouse Chromosome 11 with LOD=4.3. This locus is named Tgq22 (triglyceride QTL 22). Tgq22 is also linked to plasma triglycerides after 3 weekson a high fat diet at 19 cM (LOD=5.4). The peak marker for Tgq22 is rs3023254 (38 Mb). DBA/2J-derived alleles at Tgq22 confer increase triglyceride concentrations with a stronger effect in males compared to females. The QTL confidence interval spans 11cM to 26 cM. |
