Mutation details: This locus can generate two alternatively spliced variants that encode either a lysosomal sialic acid acetylesterase (Lse) or a cytosolic esterase (Cse). Exon 2 is unique to the Lse transcript, encoding the N-terminus of the protein product. A floxed neomycin resistance cassette was placed upstream of exon 2 and an additional loxP site was placed downstream of exon 2. Transient cre expression in correctly targeted ES cells removed the neomycin selection cassette leaving exon 2 flanked by loxP sites.
