Genetic loci associated with lipopolysaccharide (LPS) induced B-cell activation were mapped using 139 animals from a (C57BL/6Igc x BALB/cIgc)F2 intercross. Parental strain C57BL/6Igc displays higher B-cell activation after LPS stimulation (as measured byCD69 expression in CD19+ splenocytes) compared to parental strain BALB/cIgc. The F2 mice were genotyped for 82 polymorphic markers at an average resolution of 20 cM.

Significant linkage to LPS-induced B-cell activation mapped to 20.4 cM on mouse Chromosome 17 near D17Mit24 (LOD=3.4). This locus is in close linkage with the H2 locus and is named Lbca1 (LPS-induced B cell activation 1). C57BL/6Igc-derived alleles at Lbca1 confer increased B-cell response with recessive inheritance. Reciprocal congenic lines were used to confirm the presence of Lbca1. First, congenic line B6.C-H2d/bByJ carries BALB/c-derived DNA from 18.7 cM to 20.43 cM on chromosome 17 on a C57BL/6 genetic background. These animals displayed decreased CD69 expression, decreased proliferation, and decreased IgM secretion after LPS-stimulation compared to background strain C57BL/6. Second, C.B10-H2b/LilMcdJ congenic animals carry C57BL/10-derived DNA at the same genetic interval on chromosome 17 on a BALB/c genetic background. These animals display increased CD69 expression, increased proliferation, and increased IgM secretion after LPS stimulation compared to BALB/c animals. These findings are consistent with an H2-linked locus, namely Lbca1, influencing the LPS response of B-cells.

Suggestive linkage to LPS-induced B-cell activation mapped to 42.5 cM on mouse Chromosome 4 near D4Mit301 (LOD=2.7). C57BL/6Igc-derived alleles at this locus confer increased B-cell activation with an additive mode of inheritance. This locus is tentatively designated Lbca2 (LPS-induced B cell activation 2). Tlr4 at 33 cM is a potential candidate gene for Lbca2.
