A series of genome-tagged congenic lines derived from CAST/EiJ donor DNA segments on a C57BL/6J genetic background were used to map genetic loci involved in noise-induced hearing loss. The genome-tagged mice consist of 39 chromosome-specific lines covering approximately 80%of the genome. Animals were exposed to 108 dB sound pressure level (SPL) intensity noise at 6-8 weeks of age and assessed for hearing 23 days later via auditory brainstem response (ABR). Parental strain C57BL/6J is susceptible to noise-induced hearing loss whereas parental strain CAST/EiJ is resistant. Pre-exposure thresholds were similar between parental strains.

Genome-tagged congenic line B6.CAST.4PM displayed permanent threshold shift at one-third the level of parental strain C57BL/6J. B6.CAST.4PM carries CAST/EiJ-derived donor DNA from 0 cM to 67 cM (between rs13477534 and rs6355453) which localizes a resistance QTL for noise-induced hearing loss to this interval. CAST/EiJ alleles at this locus confer enhanced recovery from initial hearing damage and the QTL is named Nihlr1 (noise-induced hearing loss resistance 1). Animals from congenic line B6.CAST.4PM display almost complete recovery 23 days after noise-induced damage. Fbxo2 at 147.5 Mb (71 cM) is located near the Nihlr1 interval and is potentially a candidate gene. More analysis is required to identify candidate genes as the QTL interval is large.

Genome-tagged congenic line B6.CAST.2P displayed decreased recovery from noise-induced hearing loss compared to C57BL/6J. Thiscongenic line carries CAST/EiJ-derived alleles at the proximal portion of chromosome 2 on a C57BL/6J genetic background indicating this region confers susceptibility to initial noise-induced hearing damage. This QTL is designated Nihls1 (noise-induced hearing loss susceptibility 1). Animals from B6.CAST.2P congenic line displays minimal recovery 23 days after noise-induced damage. The precise map location for the CAST/EiJ-derived congenic interval in B6.CAST.2P is not specified.
