Mutation details: A loxP-neo-loxP cassette was inserted in a PstI site upstream of exon 3, and a second loxP site was inserted in an EcoRI site upstream of exon 1. Correct targeting was confirmed by Southern blotting. Supernatant analysis of activated homozygote macrophages demonstrated secretion of protein product was less than 25% of wild-type controls. Similar ELISA analysis demonstrated a decrease in MCP-3 secretion, which is encoded by the nearby Ccl7 gene. This is considered a hypomorph allele.
