Mutation details: Sequence encoding the transmembrane and cytoplasmic regions of the human HLA class I alpha chain was positioned in frame adjacent to exon 15 to replace the analogous region of the endogenous gene product. A floxed neomycin selection gene and an IRES-GFP followed by a stop codon were inserted downstream before exon 16. Founder mice were crossed with cre deleter mice to remove the neomycin cassette. GFP expression was detected in B cells and follicular dendritic cells. The gene fusion product's expression levels were comparable to levels of the endogenous receptor in wild-type mice. The gene fusion product retained the ability to bind the complement factor C3d but lost the ability to form a complex with CD19.
