Mutation detailsA DNA fragment from an intron of the Myh6 gene containing the micro RNA coding sequence was replaced with a floxed neo cassette via homologous recombination. Cre mediated recombination then removed the neo cassette leaving a single loxP site in the intron. PCR analysis confirmed the splice pattern of Myh6 was not disturbed. Northern blot analysis confirmed the absence of micro RNA expression in hearts from homozygous mice. (J:152696)
