Mutation detailsA targeting vector was designed to insert a viral 2A oligopeptide that mediates ribosomal skipping and a Cre recombinase gene immediately downstream of the parvalbumin translational STOP codon. The targeting vector contained, from 5' to 3', a 3 amino acid linker sequence, an frt3 site, a viral 2A oligopeptide, a Cre recombinase gene, a translational STOP sequence (TAA codon), an attB site, a PGK-Neo-polyA cassette, an frt5 site, an RNA splice acceptor, the 3' portion of the hygromycin gene with SV40 polyA signal, and attP site. This construct was electroporated into (129S6/SvEvTac x C57BL/6)F1-derived G4 embryonic stem (ES) cells. (J:155793)
