Mutation details: These MHCminTetO-Capn1 transgenic mice were generated by inserting the entire coding region of the mouse Calpain 1 gene into an MHCminTetO-promoter construct. The expression of this construct was driven by a mouse alpha-myosin heavy chain (alpha-MHC; Myh6) promoter under control of a tetracycline operator (tetO), followed by a human growth hormone polyadenylation (polyA) signal. This construct also contained 3 altered GATA sites and 2 altered thyroid-like response elements (TRE) maintaining low levels of cardiac-specific expression and decreased background. Three lines were created (L1, L2, and L3) with ascending levels of expression.
