Mutation details: A transgenic construct containing the rtTA gene under the control of the human lung-specific SFTPC promoter and intronic and polyadenylation sequences from the human GH gene, and another transgenic construct containing the murine Tnf gene under the control of TRE (tetO), were co-injected into fertilized mouse eggs to generate transgenic mice in which the two constructs were integrated at the same chromosomal location. Line 320-1 was extensively investigated. Northern blot analysis showed that Tnf mRNA levels were significantly increased in the lung, and no Tnf transgene message was observed in the heart, liver, kidney, muscle, spleen, or thymus; however, a weak band was observed in the brain after doxycycline treatment. TNF activity was significantly increased about 6-fold in the lungs of doxycycline-treated transgenic mice.
