Mutation detailsA targeting vector was generated that contained 5 + 3' homology arms to the first intron of the Gt(ROSA)26Sor locus, a cassette containing a splice acceptor, the improved rtTA-M2 element followed by a human TP53 intron and in opposite orientation, the tet-response element TRE-tight promoter driving a cre coding sequence (tetO-cre). The vector also contained an FRT-flanked neo selection cassette. Homologous recombination was done in R1 ES cells. Germline mice were produced and subsequently crossed to Flpe deleter mice to delete the neo cassette. (J:159515)
