Mutation details: A BAC containing the Runx2 locus was modified through ET recombination to insert a codon-improved cre recombinase sequence and an ampicillin resistance gene flanked by two FRT sites at the translational start site of the distal promoter (P1) of the Runx2 gene. Subsequently in the recombinant BAC, the ampicillin resistance cassette was removed by arabinose-induced FLP recombination.The linearized construct was injected into the pronuclei of FVB/N oocytes. Three founder lines were established and one was chosen for experimental use based on its expression pattern.
