Mutation details7 kb of the 5' UTR of the human ATP6V1B1 gene (B1 promoter region) was cloned upstream of cre recombinase in the construct. The purified transgene fragment was injected into single-cell embryos. 50 founders were produced, and line 45 was analyzed further. Founders were bred onto a C57BL/6 x CBA background. Cre activity was assessed by crossing cre transgenic animals to mice carrying a floxed fluorescent reporter. Activity was observed in all intercalated cells in the collecting duct and in most cells of the connecting segment (including about half of the principal cells) and in the male reproductive tract, while low levels of activity were identified in non-ATP6V1B1 expressing tissues including the brain, large and small intestine, and uterus. No expression was detected in liver, stomach, skeletal muscle, heart or spleen. (J:162708)
