Mutation details: A vector containing a cre-loxP-neo-loxP cassette flanked by two short homologous sequences was electroporated into the bacteria (EL250) together with a modified BAC (RP23-16G10) containing the Mcpt8 locus. By homologous recombination, the coding sequence of cre recombinase was inserted directly behind the Mcpt8 start codon. The neo cassette was deleted by growing bacteria in arabinose-containing medium and the BAC clone was injected into the pro-nucleus of fertilized C57BL/6 oocytes. The copy number of the integrated BAC in two different founder lines was determined by quantitative PCR for Mcpt8 (5 copies in line #1 and 7 copies in line #2.)
