Mutation detailsA transgenic construct was generated with the En1 cDNA driven by a 520 bp apical ectodermal ridge (AER)-specific Msx2 promoter. The purified construct was injected into Swiss Webster zygotes. Six founders were obtained. Founder 86 showed a dramatic limb phenotye, while 2 others (25, 95) appeared normal, but offspring had similar phenotypes (line 25; 100% of pups and line 95; 60% of pups) seen in line 86 (100% of pups). Two founders did not transmit the transgene, while the other transmitted the transgene but offspring showed no phenotype. Line 86 was studied in detail. (J:62572)
