• Mutation details: Most of intron 10 and all of exon 11, encoding the linker domain and 5' terminus of the POUh domain, were replaced with a neo cassette via homologous recombination. RT-PCR detected the presence of a mutant transcript with exon 10 spliced to exon 12. EMSA analysis with nuclear extracts from spleen and thymus cells from heterozygous mice indicates that any truncated protein made is not able to bind DNA. This suggests that this mutation results in a null allele. (J:165572)
