Mutation detailsThe BAC RP23-20N4 containing the entire Spo11 locus (including 188kb of sequence upstream of the initiation codon and 57kb downstream of the stop codon) was engineered by homologous recombination to insert an IRES-cre-(Frt-flanked) neo fragment immediately downstream of the stop codon. The neo was subsequently removed by Flp in bacteria. The modified BAC was injected into the pronuclei of fertilized eggs. Two named lines, D5 and H9, were generated and studied further. (J:166888)
