Mutation details: A loxP site was inserted at the end of intron 4 and a neo cassette flanked by two loxP sites were inserted into the middle of intron 6. The neo cassette was deleted by transiently transfection of an expression vector of Cre recombinase in the ES cells. Excision of the fifth exon and sixth exon by Cre recombinase deletes part of the tyrosine kinase domain and fully inactivate the gene as determined by Western blot, creating a null allele.
