Mutation detailsA fragment containing the cre/ERT2 sequence was inserted into a plasmid containing the human DCX promoter. A 7.7 kb fragment of the 3' UTR of DCX was generated by PCR and inserted into the fragment. The construct was injected into FVB fertilized oocytes. Two founders were obtained, with both displaying the expected recombination pattern with tamoxifen treatment after crossing to reporter mice. Southern blot analysis suggested only 1 copy of the transgene had been integrated into the genome. Since activity could be detected in line 1 with no tamoxifen treatment, only line 2 was propagated further. (J:167201)
