Mutation details: BAC recombineering was used to generate a vector with the cre/ERT2 sequence fused with the start codon in exon 2 of a partial advillin gene sequence located in BAC clone RPCI23-424F19. An Frt-flanked kanamycin selection marker was included in the initial cre cassette and was subsequently excised by induction of Flp recombinase in EL250 cells. The purified construct was used for pronuclear injection to generate 8 founder lines. Cre activity was verified in offspring of founders by breeding to ROSA26-lacZ reporter mice. Founder A exhibited strongest staining in DRGs and was analyzed in detail. Line A carries about 10 transgene copies (estimated by Southern blotting.) The transgene has integrated into the intronic region between exons 10 and 11 of the Ccser1 gene on chromosome 6 (chr6: 62276099-62638573). The insertion creates an 82 bp deletion.
