Mutation details: This transgene consists of the SV40 early genes including the coding sequence of the transforming large tumor and small tumor antigens under the control of the rat L-type pyruvate kinase gene regulatory region. Transcription of Pklr is dependent on metabolic and hormonal controls such that glucose and insulin increase transcription while fasting, refeeding a low-carbohydrate diet and glucagon inhibit Pklr transcription. Thus manipulation of diet can result in expression of the transgene. Three lines were established with different transgene copy number (Lines 1,2,3). Line 1 carries approximately 100 copies of the transgene. Northern blot analysis indicates transgene expression in the liver and to a lesser degree in the small intestine, kidney, and pancreas.
