Mutation details: An Frt-flanked zeomycin-puromycin selection cassette was inserted after an IRES-cre-IRES-EGFP sequence to generate a construct for homologous recombination. The construct was inserted after the stop codon of the Gucy2g (GC-G) gene in the bacterial artificial chromosome BAC RP23-345B13. expression of flippase in EL250 bacterial cells removed the selection cassette. The resulting modified BAC construct was injected into fertilized pronuclei to generated transgenic mice. Eight founders were obtained, with seven lines showing transgene expression in the Grueneberg ganglion. Line 52 exhibited robust GFP expression and was selected for further analysis.
