Mutation details: The construct was constructed by inserting a cassette containing the FLPe/ERT2 coding and polyA sequences (enhanced FLP fused to a triple mutant estrogen receptor) into a vector containing the CAG promoter (chicken beta-actin promoter coupled to CMV early enhancer sequence). Tandem insulator sequences that include the chicken beta globin hypersensitive sites flanked the entire construct. The purified transgene was injected into fertilized eggs to produce transgenic animals. Eight lines were obtained. After treatment with tamoxifen, all showed ability to recombine and activate the R26::FRAP reporter when crossed to mice carrying that allele. The line showing the greatest induction of alkaline phophatase activity in embryos was characterized in further detail. Cre activity was found to be low in most adult tissues and not effective in the spleen or skin (J:186310).
