Mutation details: A BAC containing the resistin (Retn) gene was modified by inserting a fragmemt encoding a nuclear localization signal (NLS) sequence, cre recombinase, and a polyadenylation signal in front of the start codon of Retn. Start sites of other genes in the BAC were mutated to prevent aberrant transcription. The purified construct was injected into fertilized C57BL/6 oocytes and founders were produced. These were crossed to wild-type and the mice were mated to ROSA26R reporter mice to assess cre activity. The line exhibiting the greatest specificity and efficiency was used for subsequent analysis.
