Mutation details: A human beta-globin locus control region HS2 (DNaseI hypersensitive site 2) fragment was linked upstream in tandem to an HBB transgene that has a lacZ cassette in place of the region from the initiation codon to the BamH1 site of the HBB gene sequence. Regulatory sequences of HBB are retained in the 5' flanking region, in IVS-2 (second intervening sequence), and 3' to the gene. 10 founder lines were produced, and line A (copy number of 57) which has high beta-galactosidase activity in the periperal blood. The ES cell line generated from this transgene carries the transgene on distal chromosome 4.
