Mutation detailsThis transgene consists of a 36.8-kb SwaI fragment of the BAC clone RP23-551L11 (from 129S6/SvEvTac) that contains the complete Wnk4 transcription unit plus 9 kb of 5' flanking and 11.5 kb of 3' flanking sequence. A synonymous substitution at codon 524, creating a new AvaI cleavage site was introduced. Fluorescence in situ hybridization showed a single site of insertion. Transgene expression was detected in heart, kidney, testis, brain, and lung but not in liver or spleen. RT-PCR showed that total amount of mRNA is increased by about 2-fold.
