Mutation details: The mutagenic insertion and chromosome engineering resource (MICER) clone MHPP322c09 was used to insert a loxP site at the 3' end of the keratin type II cluster, downstream of the type I Keratin 18 (Krt18) gene. This clone contains a 6.9 kb insert spanning 101,876,016-101,882,964 bp on chromosome 15 (genome build unknown) and includes a puromycin resistance gene cassette, a loxP site, an HPRT mini-gene, and the agouti (A) coat color gene. The construct was transfected into AB2.2 ES cells carrying Krt1ctm1Tmm and double-targeted clones in cis were identified for blastocyst injection. Cre-mediated removal of both loxP sites can be used to delete all of the type II keratins and one keratin type I, Krt18.
